The catecholamine-induced increase in contractile force in mammalian myocardium is mainly due to activation of beta adrenoceptors. The existence of myocardial alpha-1 adrenoceptors mediating positive inotropic effects is, however, now well established Osnes et al., 1985; Fedida 1993; Terzic et al., 1993 ; . In rat and rabbit heart the alpha-1 adrenoceptormediated effect is influencing and contributing to the final inotropic effect of combined receptor activation by the endogenous agonist norepinephrine Skomedal et al., 1988, 1990a ; . As alpha-1 adrenoceptor-mediated and beta adrenoceptormediated inotropic effects can be differentially influenced Endoh and Motomura, 1979; Christiansen et al., 1987 ; , it has been speculated that alpha-1 adrenoceptor mediated effects may have increased importance during conditions where beta-adrenoceptor mediated effects are attenuated Christiansen et al., 1987, Skomedal et al., 1988, 1990a ; . What started as a simple plan to share some print resources has developed into a much deeper and more meaningful program as we explore the rich resources we both have to share with each other. We are now exchanging expertise and setting up a modern Pen Pals program for children from Pickering's after-school program and the Georgina Island elementary school. Pickering plans to launch a full program for National Aboriginal Day 2005 with guidance and support from the Chippewas of Georgina Island. We hope our project model can contribute to raising general awareness of Ontario's First Nation peoples, communities, and libraries while shedding some light, for First Nation peoples, on their non-native colleagues and neighbours.

Cross-reactivity is commonly encountered in immunoassays 1-4, 10 ; and was evident here. There appears to be a relationship between drug structure and cross-reactivity. In the case of the barbiturates, amobarbital and pentobarbital cross-reactivities were similar in comparison to secobarbital ` to 1 whereas phenobarbital substitution of a phenyl group at the 5 position ; and thiopental sulfur substitution within the ring ; were much less reactive. A nonbarbiturate sedative-hypnotic, gluthethimide, and a series of narcotic analgesics, amphetamine and methamphetamine, were essentially noncrossreactive in the barbiturate radioimmunoassay.

Fluorescence has many advantages over traditional colour and radioactive labels, and is playing an increasingly important role in the most powerful analytical techniques. For example, fluorescence is at the heart of many nucleic acid based diagnostics e.g. DNA microarray, real time-PCR, fluorescence in situ hybridisation, etc ; , immunofluorescence assays, defined substrate technologies and differential display proteomics and is gradually replacing or complementing other techniques based on colour or radiolabels. There are several reasons for this: Fluorescent techniques are 106 x more sensitive than those based on colour and suffer minimal interference. Fluorescence signals are proportional to the concentration of the substance being investigated. Fluorescencebased measurements are readily quantifiable over many orders of magnitude compared with two orders of magnitude for colour-based assays. Fluorescence is non-invasive and changes in fluorescence intensity can be measured remotely and very rapidly in picoseconds if necessary ; . This allows the study of dynamic processes, in real-time, such as rapid physiological changes inside a cell. Fluorescence techniques are generally simple and safe compared to similarly sensitive radioactive methods, which create disposal problems and require specialised facilities and training. Fluorescence is detected and quantified using a variety of instrumentation such as fluorescence microscopes, scanners, plate readers, fluorometers as well as flow cytometers. Despite the widespread use of fluorescence techniques, only a limited number of fluorochrome families are known Table 1 ; . Thus there is a need for new colours of fluorescent labels that can be used to complement existing fluorophores in multiplex assays.

For detection of salmonella at genus level, inv-A universal primer was selected. In order to identification of Salmonella typhimurium, specific primers of Rfbj, Fljb and Flic related on gene sequences of O4, H2: 1, 2 and H1: i respectively were used. Results: Of the 400 fecal samples tested in this study, 33 8.25% ; was culture positive for Salmonella serotypes. Of these, 22 of 33 66.7% ; isolates were identified as Salmonella typhimurium. All 22 cultured positive samples, also were positive by m - PCR. The sensitivity and specificity both methods were 100%. Conclusion: Our results showed using m PCR by selected specific primers could be achieved easily for Salmonella typhimurium detection and results had been confirmed effortlessly within a 8 hours compared to 7-8 days for the conventional microbiological methods.

LOWE * Department of Clinical Chemistry, Intensive Care Unit, Clinical Audit Department, Fazakerley Hospital, Liverpool L9 7AL Mortality in the Intensive Care Unit ICU ; is approximately 30%, half of whom die of sepsis and multiple system organ failure MSOF ; .1 Microalbuminuria has been shown to be a non-specific but sensitive marker of systemic inflammation with a more rapid onset than other common inflammatory markers.2 We investigated the use of microalbuminuria as an indicator of the development of sepsis and MSOF and compared this with other acute phase markers C-reactive protein and erythrocyte sedimentation rate ; over three days. Fifty critically ill patients in the ICU were recruited to the study after informed agreement from relatives. Blood samples were collected upon admission and daily for up to three days for the determination of C-reactive protein CRP ; and erythrocyte sedimentation rate ESR ; by immunoturbidimetry3 and the Westergren technique4 respectively. Urine samples were collected every 6 h for 18 h and the microalbumin: creatinine ratio determined by immunoturbidimetry and Jaff reaction.5 To assess the progress of organ dysfunction, multiple organ dysfunction scores MODS ; were obtained daily up to a maximum of 10 days.6 For statistical analysis, MannWhitney and chi-square tests were used as appropriate. Outcome was assessed according to patient survival survivors, n: 36; non-survivors, n: 14 ; . ESR and CRP values obtained throughout the study did not show any association with outcome in either group. Microalbumin: creatinine ratios reference range : 3.5 mg: mmol ; obtained within 612 h of admission were significantly different survivors: median m ; : 3.3, interquartile range r ; : 114; non-survivors: m: 22, r: 354; p: 0.01 ; . Microalbuminuria concentrations were in concordance with the MODS on day 1 p: 0.002 ; . We concluded that micoralbuminuria measurements can be used to identify high-risk patients, the earlier identification of these patients means that they can be targeted more accurately with more aggressive clinical intervention.
