With the steady increase in life expectancy cancer continues to grow into a major health problem. An estimated 35% of human cancer-related mortality is attributed to diet 1 ; . While the diet is a source of carcinogens it is also the source of inhibitors of the carcinogenic process. Laboratory based research over the past few decades has established an important role for fruits and vegetables as cancer risk reducers either by blocking cancer initiation or suppressing promotion 2 ; . More recently several groups have shown that phytochemicals present in spices also specifically target pathways that are deregulated in cancer. Since multiple pathways are deregulated in cancer the consumption of a variety of plant products improves the chances of targeting multiple deregulated pathways in cancer. For example compounds such as eugenol from cloves and bay leaves ; , curcumin turmeric ; , capsaicin peppers ; , gingerol ginger ; , and diallyl sulfide garlic ; have been found to regulate specific pathways involved in cell cycle progression, apoptosis, survival, intracellular signaling, as well as cellular detoxification 3 ; . Melanoma Melanoma is the deadliest form of skin cancer accounting for almost 78% of skin cancer fatality. It is one of the fastest growing cancers in the developing world. The 5-year recurrence rates of patients with high-risk melanoma are 5580% and survival rates are poor 4 ; . Adjuvant therapy using interferon given to stages II, III, and IV following surgery produce a response rate of 1020% but only for a short duration 3 9 ; . Chemotherapeutic agents such as dacarbazine DTIC ; show a response rate of 1520% and a combination of cisplatin and dacarbazine with tamoxifen and carmustine produce a response rate of 55% 4 10 ; . However, the typical short life of the response rates has had no significant effect on overall survival. Ongoing and completed clinical trials using monoclonal antibodies and vaccine therapy for malignant melanoma treatment have been promising but overall have not made an impact on malignant melanoma mortality rates. Hormonal therapy of disseminated melanoma has been used infrequently and is generally regarded as being ineffective, although use of NSC-17256 a drug with glucocorticoid and anti-estrogenic activities ; produced objective tumor regression in 5 of patients 5 11 ; . Dietary Agents for Melanoma Chemoprevention Thus far, not many dietary agents have been tested for chemopreventive activities against malignant melanoma although there are many potential agents that target specific pathways deregulated during the development of malignant melanoma. In almost all these cases provocative preclinical data has been provided but no randomized trials have been undertaken to carefully evaluate the pathways targeted, establish dose-response relationships, validation of biomarkers, etc. Use of Eugenol as a Botanical Chemopreventive Agent 4-allyl-2-methoxyphenol, also known as Eugenol, is found in reasonable quantities in the essential oils of different spices such as Syzgium aromaticum clove ; , Pimenta racemosa bay leaves ; , and Cinnamomum verum cinnamon leaf ; . These spices are used as flavoring agents in culinary practices all over the world. Further, eugenol has been used as an antiseptic, anti-bacterial, analgesic agent in traditional medical practices in Asia. We have shown that eugenol exhibits potent anti-proliferation activity in melanoma cells that represent different stages of progression. We have also shown that eugenol blocks cells in the S phase of progression; induces apoptosis; and inhibits E2F1 transcriptional activity in melanoma cells. Further overexpression of E2F1 was able to overcome the growth inhibition mediated by eugenol 5 ; . Eugenol also inhibited the invasion of melanoma cells through the basement membrane in Boyden chamber assays. In the B16 xenograft model intraperitoneal delivery of eugenol inhibited melanoma metastasis to the lungs compared to untreated controls. These results suggest that it is possible to use dietary agents and botanicals for the inhibition of malignant melanoma. In the case of melanoma, a better approach may be to screen such phytochemicals in a secondary prevention setting to prevent the progression of easily treatable primary disease to the highly aggressive metastatic disease that is extremely resistant to current therapeutic regimens.

EFFECT ON CEREBRAL VASCULAR RESISTANCE AND INCORPORATION INTO CEREBRAL ADENINE NUCLEOTIDES By Robert M. Berne. Rafael Rubio, and Richard R. Curnish ABSTRACT A threefold increase in tissue adenosine levels was produced by ischemia in the dog and the rat brain within 1 minute; adenosine levels increased further with longer periods of ischemia. Inosine and hypoxanthine were also increased by ischemia but to different degrees. The nucleosides and hypoxanthine appeared in cerebrospinal fluid during ischemia in the dog brain, and incubation of adenosine in normal cerebrospinal fluid failed to show the presence of degradative enzymes. Intra-arterially administered adenosine in the dog and the cat produced little or no increase in cerebral blood flow or the diameter of pial arterioles, respectively, when it was given in amounts that reduced arterial blood pressure. However, when it was applied topically to exposed pial arterioles of the cat, adenosine induced dilation that was roughly proportional to the dose used. When U-MC-adenosine was infused into the internal carotid arteries of the dog, no radioactivity was detectable in the cerebrospinal fluid and practically none appeared in the brain tissue. When labeled adenosine was added to dog cerebrospinal fluid, only a few counts appeared in the cerebral venous blood, whereas cerebral tissue was heavily labeled with 84-87% of the radioactivity in the form of adenine nucleotides. In the rat, after intravenous injection of labeled adenosine, the counts per gram of heart were twenty- to thirtyfold greater than those per gram of brain. These observations indicate that intra-arterially administered adenosine probably fails to cross the blood-brain barrier rapidly enough to influence cerebral blood flow but that it can be released from the ischemic brain into the cerebrospinal fluid and be reincorporated from the cerebrospinal fluid into brain nucleotides. Hence, adenosine can conceivably participate in the regulation of cerebral blood flow.
